Antibody F(ab)₂ fragmentation kits

Description: The F(ab)₂ Fragmentation kits are designed for the generation and isolation of F(ab)₂ fragments from IgG molecules. The kit utilises Immobilised Pepsin resin. Pepsin is a proteolytic enzyme that is routinely used for the generation of F(ab)₂ fragments from immunoglobulin G (IgG). One or more of the disulfide bonds that join the heavy chains in the hinge region are preserved, so the two Fab regions of the antibody remain joined together, yielding a divalent molecule (containing two antibody binding sites), hence the designation F(ab)₂. The light chains remain intact and attached to the heavy chain, whereas the Fc fragment is digested into small peptides. The Immobilised Pepsin offers the distinct advantage of eliminating enzyme contamination of the F(ab)₂ fragments.

Description: Le kit d'ARN total E.Z.N.A.® MicroElute fournit une méthode rapide et simple pour isoler jusqu'à 50 µg d'ARN total à partir de petites quantités de cellules eucaryotes cultivées, de tissus tels que les échantillons disséqués au laser (LDS) ou les aspirats à aiguille fine (FNA). Normalement, jusqu'à 5×10⁵ cellules eucaryotes ou 5 mg de tissus peuvent être utilisés au cours d'une même expérience, selon le type de tissu utilisé. Ce kit permet le traitement d'échantillons uniques ou multiples en moins de 30 minutes. Il n'est pas nécessaire de recourir aux extractions au phénol/chloroforme, ni aux étapes fastidieuses telles que l'ultracentrifugation en gradient de CsCI et la précipitation à l'isopropanol ou au LiCl. L'ARN purifié peut être élué avec 10 à 15 µl d'eau sans nucléase. L'ARN purifié est prêt pour la plupart des applications en aval, telles que la RT-PCR, le transfert Northern, la purification Poly A+, la protection des nucléases et la translation <i>in vitro</i>.
