Description: Digoxigenin (DIG) is a steroid found exclusively in the flowers and leaves of the plants Digitalis purpurea and Digitalis lanata. DIG is the steroid component of digoxin, a cardiac glycoside, and can be conjugated onto a variety of biomolecules (proteins, nucleic acids) to be detected in further assays (e.g. ELISA). These antibodies can be labeled with dyes, enzymes or fluorescence, directly or secondarily, for visualization and detection.
DIG has an advantage over biotin due to the fact that biotin is an endogenous molecule of living cells and plays a key role in many reactions, which can sometimes lead to false positive results. Because the blossoms and leaves of Digitalis plants are the only natural sources of digoxigenin, binding of the anti-digoxigenin antibody in other biological material is not a consideration.

Description: Protein phosphorylation is a fundamental event in the regulation of a large number of intracellular processes. Phosphorylation of specific tyrosine residues is the result of activation or stimulation of their respective protein tyrosine kinases. The phosphorylated proteins can be auto-phosphorylated kinases or certain cellular protein substrates. Tyrosine-phosphorylated proteins are involved in signal transduction and in the regulation of cell proliferation. Antibody to phosphotyrosine provides an excellent tool for the detection, characterization, and purification of phosphotyrosine containing proteins. This MAb shows no cross-reaction with other phosphoamino acids and is superb for multiple applications including staining of formalin/paraffin tissues.

Description: Recognizes a phosphor-protein of 45 kDa, identified as MyoD1. The epitope of this MAb maps between amino acid 180-189 in the C-terminal of mouse MyoD1 protein. It does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Weak cytoplasmic staining is observed in several non-muscle tissues, including glandular epithelium and also in rhabdomyosarcomas, neuroblastomas, Ewing s sarcomas and alveolar soft part sarcomas.

Description: MAb VU-2G7 reacts with MUC1, a large transmembrane glycoprotein expressed on the ductal surface of normal glandular epithelia. The dominant epitope of VU-2G7 has not been established with epitope fingerprinting. The extracellular domain of MUC1 largely consists of a highly conserved, O-glycosylated 20 amino acids tandem repeat which can occur 30-100 times per molecule depending on the length of the allele involved. In the vast majority of human carcinomas this protein is up regulated and poorly glycosylated and appears on the cell surface in a non-polarized fashion.

Description: This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It is reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

Description: Recognizes a 77-85 kDa protein, identified as cellular or tissue transglutaminase II (TGase II). Transglutaminases are enzymes that catalyze the crosslinking of proteins by epsilon-gamma glutamyl lysine isopeptide bonds. While the primary structure of transglutaminases is not conserved, they all have the same amino acid sequence at their active sites and their activity is calcium-dependent. The protein encoded by this gene acts as a monomer, is induced by retinoic acid, and appears to be involved in apoptosis. Finally, the encoded protein is the autoantigen implicated in celiac disease. The identification of transglutaminase as the main antigen of endomysium antibodies allows a new diagnostic approach to celiac disease (CD), a genetic, immunologically mediated small bowel enteropathy that causes malabsorption. TGase II is implicated in programmed cell death, signal transduction, drug-resistance, cell growth, endocytosis, insulin secretion, cell adhesion, cataract formation, and wound healing.

Description: Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes. The regulatory domain of MyoD is approximately 70 amino acids in length and includes both a basic DNA binding motif and a bHLH dimerization motif. MyoD family members share about 80% amino acid homology in their bHLH motifs. Anti-myogenin labels the nuclei of myoblasts in developing muscle tissue, and is expressed in tumor cell nuclei of rhabdomyosarcoma and some leiomyosarcomas. Positive nuclear staining may occur in Wilms' tumor.
