Description: Ankyrins are membrane adaptor molecules that play important roles in coupling integral membrane proteins to the spectrin-based cytoskeleton network. Mutations of ankyrin genes can lead to severe genetic diseases, such as fatal cardiac arrhythmias and hereditary spherocytosis. ANKRD13B (ankyrin repeat domain 13B) is a 626 amino acid protein that contains two ANK repeats and three ubiquitin-interacting motif (UIM) repeats. Conserved in dog, cow, mouse and rat, ANKRD13B exists as two alternatively spliced isoforms. The gene that encodes ANKRD13B maps to human chromosome 17, which makes up over 2.5% of the human genome, with about 81 million bases encoding over 1200 genes. Two key tumour suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. BRCA1 is recognised as a genetic determinant of early onset breast cancer. Chromosome 17 is also linked to neurofibromatosis, dysregulated Schwann cell growth, Alexander disease, Birt-Hogg-Dube syndrome and Canavan disease.

Description: The deduced 257 amino acid protein PACRG (Parkin co-regulated gene) shows potential links to the ubiquitin/proteasome system. PACRG and Parkin are attached in a head-to-head arrangement on opposite DNA strands and share a common 5' flanking promoter region. The PACRG gene maps to chromosome 6q26; Northern blot analysis detects PACRG expression in all tissues examined except placenta. Using a positional cloning strategy in 197 Vietnamese leprosy simplex families (i.e. families with two unaffected parents and one affected child), significant connections between leprosy and 17 markers in the 5' regulatory region that PARK2 and PACRG share were observed. Possession of two or more of the 17 risk alleles is highly predictive of leprosy.PACRG is a gene located very close to parkin, in reverse orientation on the chromosome. It is thought to be co-transcribed with parkin by a bi-directional promoter between the two genes.

Description: DERP6, is a 316 amino acid protein that localizes to the cytoplasm and exists as multiple alternatively spliced isoforms. Expressed ubiquitously with highest expression in liver, heart, testis, brain and skeletal muscle, DERP6 is thought to be involved in p53-mediated transcriptional regulation. The gene encoding DERP6 maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, though specifically it is recognized as a genetic determinant of early onset breast cancer and predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes.

Description: HEXDC, also known as hexosaminidase D, beta-hexosaminidase D, N-acetyl-beta-galactosaminidase, hexosaminidase domain-containing protein or beta-N-acetylhexosaminidase, is a 486 amino acid cytoplasmic and nuclear protein that has hexosaminidase activity and belongs to the glycosyl hydrolase 20 family. Existing as two alternatively spliced isoforms, HEXDC catalyzes the hydrolysis of non-reducing N-acetyl-D-hexosamine residues near the termini of N-acetyl-beta-D-hexosaminides. The gene encoding HEXDC maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes. Description: The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21.<be>This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. [provided by RefSeq, Aug 2008].

Description: C4orf17 (chromosome 4 open reading frame 17) is a 359 amino acid protein encoded by a gene that maps to human chromosome 4. Human chromosome 4 represents approximately 6% of the human genome and contains nearly 900 genes. Notably, the Huntingtin gene, which is found to encode an expanded glutamine tract in cases of Huntington's disease, is on chromosome 4. FGFR-3 is also encoded by a gene that is located on chromosome 4, and has been associated with thanatophoric dwarfism, achondroplasia, Muenke syndrome and bladder cancer. Chromosome 4 is also tied to Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.

Description: CD300C , also known as CLM-6 (CMRF35-like molecule 6), is a 224 amino acid single-pass type I membrane protein that is present on the surface of neutrophils, monocytes and a select proportion of peripheral blood B and T lymphocytes. CD300C contains two potential N-gylcosylation sites, a potential O-glycosylated hinge-like region and an Ig-like V-type (immunoglobulin-like) domain that is very similar to the Fc receptor for polymeric IgA and IgM. The gene encoding CD300C is localized to a region on human chromosome 17 that harbors a susceptilibty locus for psoriasis, dermatitis and rheumatoid arthritis, suggesting a possible involvement of CD300C with these conditions.

Description: The 3-HAO family of proteins consists of intramolecular dioxygenases that contain non-heme ferrous iron. HAAO (3-hydroxyanthranilate 3,4-dioxygenase), also known as HAO or 3-HAO, is a 286 amino acid monomeric cytoplasmic protein that belongs to the 3-HAO family. Utilizing Fe²⁺ ions as a cofactor, HAAO participates in the formation of quinolinic acid (QUIN), and excitotoxin, by catalyzing the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde. HAAO may play a role in disorders that are associated with varying levels of QUIN expression, and is widely expressed in peripheral organs, including kidney and liver. Existing as two alternatively spliced isoforms, the gene encoding HAAO maps to human chromosome 2p21 and mouse chromosome 17 E4.
