In molecular cloning, vectors are DNA molecules used to artificially deliver foreign genetic material into another cell. The four major types of vectors are plasmids, viral vectors, cosmids, and artificial chromosomes, with the most commonly used of these being plasmids. Vectors may be used for cloning, and certain vectors are specifically tailored to that exact purpose, while others may be designed specifically for transcription and protein expression. Simpler transcription vectors are used to amplify their inserted genetic material.

Description: Acid sensing ion channel ASIC1 is present in brain as a 4.3-kb transcript with localization to rat dorsal root ganglia. In situ hybridization of rat brain suggests that ASIC1 is most abundant in the main olfactory bulb, cerebral cortex, hippocampal formation, habenula, basolateral amygdaloid nuclei and cerebellum. ASIC1 and H+-gated currents may contribute to the development of fear and anxiety. ASIC2, also designated amiloride-sensitive cation channel 1, neuronal (ACCN1), mammalian degenerin, BNAC1 (MDEG) and brain Na+ channel 1, mediates the normal detection of light touch. ASIC2 mRNA is abundant in brain, specifically in neurons. ASIC2 is expressed as 2.7- and 3.7-kb transcripts in brain and spinal cord tissues. ASIC3, also designated SLNAC1 and TNaC1, mediates detection of lasting pH changes and is involved in modulating moderate- to high-intensity pain sensation. ASIC4, also designated ACCN4 and BNAC4, is abundant in pituitary gland and is also present in the inner ear.

Description: Two members of the human zinc finger Kruppel family, ZNF 12 (KOX 3) and ZNF 26 (KOX 20), have been localized by somatic cell hybrid analysis and in situ chromosomal hybridization. The presence of individual human zinc finger genes in mouse-human hybrid DNAs was correlated with the presence of specific human chromosomes or regions of chromosomes in the corresponding cell hybrids. Analysis of such mouse-human hybrid DNAs assigned the ZNF 12 (KOX 3) gene to chromosome region 7p.

Description: Green fluorescence protein (GFP) is a 27 kDa protein derived from the jellyfish Aequorea victoria, which emits green light (emission peak at a wavelenth of 509 nm) when excited by blue light (excitation peak at a wavelenth of 395 nm). Green Fluorescent Protein (GFP) has become an invaluable tool in cell biology research, since its intrinsic fluorescence can be visualized in living cells. GFP fluorescence is stable under fixation conditions and suitable for a variety of applications. GFP has been widely used as a reporter for gene expression, enabling researchers to visualize and localize GFP-tagged proteins within living cells without the need for chemical staining. Other applications of GFP include assessment of protein protein interactions through the yeast two hybrid system and measurement of distance between proteins through fluorescence energy transfer (FRET) protocols. GFP technnology has considerably contributed to a greater understanding of cellular physiology.

Description: The ubiquitin (Ub) pathway involves three sequential enzymatic steps that facilitate the conjugation of Ub and Ub-like molecules to specific protein substrates. The first step requires the ATP-dependent activation of the Ub C-terminus and the assembly of multi-Ub chains by the Ub-activating enzyme known as the E1 component. The Ub chain is then conjugated to the Ub-conjugating enzyme (E2) to generate an intermediate Ub-E2 complex. The Ub-ligase (E3) then catalyzes the transfer of Ub from E2 to the appropriate protein substrate. A wide range of enzymes facilitate in the proteolytic Ub pathway, including monoclonal nonspecific suppressor factor-beta (MNSF-beta), a subunit of MNSF, which is a lymphokine product of a murine T cell hybrid-oma that restricts the production of LPS-induced immunoglobulin secreting cells in an antigen-nonspecific manner. MNSF-beta is a ubiquitin-like fusion protein consisting of the ribosomal protein S30 and a protein that shares 36% sequence identity with ubiquitin. This ubiquitin-like segment (Ubi-L) can be cleaved from MNSF-beta in the cytosol.

Description: iASPP (inhibitor of apoptosis stimulating protein of p53, RelA-associated inhibitor (RAI), NFKap B-interacting protein 1, protein phosphatase 1 regulatory subunit 13-like, PPP1R13L) is the third member of the ASPP family of proteins. Unlike ASPP1 and ASPP2, which interact with p53 and enhance its ability to induce apoptosis, iASPP inhibits apoptosis induced by p53 overexpression. iASPP has been identified in a yeast-two hybrid screen as an interacting protein of the p65 subunit of NFKap B (RelA),3 interacts with NFKap B in vivo, and acts as an efficient inhibitor of HIV-1 gene expression in which both NFKap b and Sp1 play major roles. iASPP expression is upregulated in human breast carcinomas expressing wild-type p53, and gene overexpression may play an important role in the leukemogenesis and/or progression of acute leukemia. Alternate Names: apoptosis stimulating protein of P53 1; Apoptosis-stimulating of p53 protein 2; Tumor suppressor p53-binding protein 2; p53-binding protein 2; p53BP2; 53BP2; Bcl2-binding protein; Bbp; Renal carcinoma antigen NY-REN-51.

Description: The adenovirus E1A protein both activates and represses gene expression to promote cellular proliferation and inhibit differentiation. CREG (cellular repressor of E1A-stimulated genes) is a cellular protein that antagonizes transcriptional activation and cellular transformation by E1A. CREG was initially isolated in a yeast two-hybrid screen due to its interaction with the TATA-binding protein, TBP. A member of the CREG family, CREG2 (cellular repressor of E1A-stimulated genes 2) is a novel protein that shares 35% homology with CREG and is expressed at highest levels in brain. CREG2 is a secreted protein containing 290 amino acids whose N-terminus is thought to function as a signal sequence. The gene encoding CREG2 maps to human chromosome 2, which consists of 237 million bases, encodes over 1400 genes and makes up approximately 8% of the human genome. A number of genetic diseases are linked to genes on chromosome 2 including Harlequin icthyosis, sitosterolemia and Alstr syndrome.
