Description: HEAT repeats form rod-like helical structures that are involved in intracellular transport. HEATR7B2 (HEAT repeat-containing protein 7B2) is a 1,585 amino acid protein that contains fifteen HEAT repeats. Existing as two alternatively spliced isoforms, the gene encoding HEATR7B2 maps to human chromosome 5p13.1 and mouse chromosome 15 A1. Chromosome 5 contains 181 million base pairs and comprises nearly 6% of the human genome. Cockayne syndrome is associated with chromosome 5 through the ERCC8 gene and familial adenomatous polyposis via the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is also chromosome 5-associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome, while deletion of the q arm or of chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.

Description: Recognizes a 60 kDa protein, identified as the heat shock protein 60 (hsp60). Its epitope is localized between aa 383-447 of human hsp60. A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp), more appropriately called as stress response proteins (srp). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone LK1, unlike LK2, recognizes only the mammalian (not bacterial) hsp60 and is useful in distinguishing hsp60 from mammals and bacteria.

Description: HEATR3 is a 680 amino acid protein existing as three alternatively spliced isoforms and containing two HEAT (Huntingtin, elongation factor 3 (EF3), protein phosphatase 2A (PP2A) and the yeast PI3-kinase Tor1) repeats. HEAT repeats form rod-like helical structures that are involved in intracellular transport. HEATR3 is encoded by a gene located on human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The GAN gene is located on chromosome 16 and, with mutation, may lead to giant axonal neuropathy, a nervous system disorder characterized by increasing malfunction with growth. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, a gastrointestinal inflammatory condition.

Description: PIRT is a 137 amino acid multi-pass membrane protein. Highly conserved among vertebrates, PIRT consists of two transmembrane domains and one putative C-terminal phosphoinositide-binding domain. Although PIRT is expressed in peripheral nervous system, with highest levels in dorsal root ganglion and trigeminal neurons, and lowest levels in sympathetic and enteric neurons, it is not expressed in spinal cord. PIRT is a required component of the VR1 complex, which positively regulates VR1, a sensor of both noxious heat and capsaicin. Correspondingly, PIRT knockout results in impaired responses to noxious heat and capsaicin exposure, while VR1 remains unaltered. The gene that encodes PIRT maps to human chromosome 17p13.1.

Description: This MAb reacts specifically with heat shock protein HSP27 in human and monkey tissues and cell lines such as MCF-7. HSP27, also referred to as the Estrogen-Regulated 24K protein and HSP28, is one of several small heat shock proteins produced by all organisms studied. HSP27 synthesis is induced by elevated temperature, as well as by estrogen in hormone responsive cells. Interestingly, human HSP27 also shares greater than 50% homology with low molecular weight Drosophila HSPs and mammalian α-crystalline lens protein. Because of the estrogen responsive nature of HSP27, this protein has been studied extensively in human estrogen responsive tissues such as cervix, endometrium and breast tissue. Therefore HSP27 may be useful in classifying various hormone sensitive tumors.
