Description: Product is the lyophilized powder of rat IgG and buffer salts.IgG antibody subtype is the most abundant serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids and provides protection from infections caused by bacteria, fungi and viruses. Maternal IgG is transferred to fetus through the placenta that is vital for immune defence of the neonate against infections. The total protein is determined using the Biuret procedure, with bovine albumin as the standard. Each vial is overfilled by approximately 10 percent so that a minimum of 20 mg of rat IgG is obtained upon reconstitution.

Description: Alpha-2-HS-glycoprotein (AHSG) is also known as Alpha-2-Z-globulin, Ba-alpha-2-glycoprotein, Fetuin-A (FETUA), is a glycoprotein that is composed of two subunits, the A and B chains, belongs to the cystatin family of proteases inhibitors and the fetuin class of plasma binding proteins. AHSG / Fetuin A is highly expressed in embryonic cells and adult hepatocytes, and is expressed to a lesser extent in monocytes/macrophages. Fetuins are carrier proteins like albumin. Fetuin-A / AHSG forms soluble complexes with calcium and phosphate and thus is a carrier of otherwise insoluble calcium phosphate. Thus AHSG / Fetuin A is a potent inhibitor of pathological calcification. Fetuin A / FETUA promotes phagocytosis of apoptotic cells and plays a role in bone development, calcium homeostasis and insulin sensitivity. Mice deficient in Fetuin-A show systemic calcification of soft tissues.

Description: Secreted Protein Acidic and Rich in Cysteine (SPARC) is a secreted, evolutionarily conserved collagen-binding glycoprotein and belongs to the SPARC family. SPARC has 286 amino acids and contains an EF-hand in C-termina domain, a follistatin-like domain with Kazal-like sequences. There are two calcium binding sites, one binds 5 - 8 Ca2+ with a low affinity and other on an EF-hand loop that binds a Ca2+ ion with a high affinity. It is highly expressed in tissues undergoing morphogenesis, remodeling and wound repair. SPARC regulate cell growth through interactions with the extracellular matrix (ECM) and cytokines. Description: beta -2-Microglobulin (B2M) is a secreted protein with 1 Ig-like C1-type (immunoglobulin-like) domain which belongs to the beta-2-microglobulin family. B2M component of major histocompatibility complex (MHC) class I molecules, involved in the presentation of peptide antigens to the immune system. Polymers of beta 2-microglobulin can be found in tissues from patients on long-term hemodialysis. B2M is a protein found on the surface of many cells and plentiful on the surface of white blood cells. Serum B2M concentration is increased in renal diseases, various malignant diseases and some inflammatory and autoimmune disorders. B2M may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. B2M has been shown as a marker for monitoring inflammatory disease activity and it appears likely to have a destructive role in amyloidosis-related arthritis. Description: The complement system provides innate defense against microbial infection and is a "complement" to antibody mediated immunity. The complement system consists of thirty five interacting plasma and membrane associated proteins which contribute to host-defense and initiate and amplify inflammation, even in the preimmune state where specific antibodies and lymphocytes are not available. In addition to the complement components themselves, this system also contains several soluble factors that prevent spontaneous complement activation from occurring in solution, as well as, several regulatory membrane associated proteins that protect host cells from accidental complement attack. Cleavage of the C3 component by either the classical pathway or the alternative pathway releases C3a and C3b. C3a appears to be important in many inflammatory responses while the C3b fragment covalently binds to the cell or bacterial surface and plays a role in opsonisation. Binding of C3b to the C4b component of the C3 convertase, results in C5 convertase (C4b3b2a) formation

The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer is standardized to an in-house standard by equivalence-point precipitation. Each IgG fraction is tested for purity and specificity at a minimum of 40 mg/mL using immunoelectrophoresis. The product is mostly goat IgG; no trace of albumin is detected.

This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. Note: F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. The product has been successfully used in Immunoblotting. It detects C3 from human, equine, hamster, Guinea pig, murine, rabbit, rat, pig, and non-human primate samples. It does not cross react wtih bovine, canine, chicken, feline and sheep samples. Description: FOXA1 is a member of the forkhead class of DNA-binding proteins. These hepatocyte nuclear factors are transcriptional activators for liver-specific transcripts such as albumin and transthyretin, and they also interact with chromatin as a pioneer factor. Similar family members in mice have roles in the regulation of metabolism and in the differentiation of the pancreas and liver.<p>FOXA1 in breast cancer is highly correlated with ERalpha+, GATA3+, and PR+ protein expression as well as endocrine signaling. FOXA1 acts as a pioneer factor for ERa in ERalpha+ breast cancer, and its expression might identify ERalpha+ cancers that undergo rapid reprogramming of ERa signaling that is associated with poor outcomes and treatment resistance. Conversely, in ERalpha− breast cancer FOXA1 is highly correlated with low-grade morphology and improved disease free survival. FOXA1 is a downstream target of GATA3 in the mammary gland. Description: TEF (thyrotroph embryonic factor) is a member of the PAR bZip (proline and acidic amino acid-rich basic leucine zipper) transcription factor family. It accumulates with robust circadian rhythms in tissues with high amplitudes of clock gene expression.Thyrotroph embryonic factor (TEF), a transcription factor, is a member of the PAR (proline and acidic amino acid-rich) subfamily of basic region/leucine zipper (bZIP) transcription factors. It is expressed in a broad range of cells and tissues in adult animals, however, during embryonic development, TEF expression appears to be restricted to the developing anterior pituitary gland, coincident with the appearance of thyroid-stimulating hormone, beta (TSHB). Indeed, TEF can bind to, and transactivate the TSHB promoter. It shows homology (in the functional domains) with other members of the PAR-bZIP subfamily of transcription factors, which include albumin D box-binding protein (DBP), human hepatic leukemia factor (HLF) and chicken vitellogenin gene-binding protein (VBP); VBP is considered the chicken homologue of TEF. Description: Poly-lysine is a polycation which binds to DNA, red cell membrane and any negatively charged protein. When adsorbed to the culture surface, poly-lysine increases the number of positively charged sites available for cell binding. A compromise between the easier to use lower molecular weight products and the extremely viscous higher molecular weights would be the products in the range of 70,000-150,000.
It is typically used as a coating substrate for culture dishes, slides, etc. It enhances electrostatic interaction between negatively charged ions of the cell membrane and the culture surface. Both the D- and L- form of the poly-lysine can be used as a coating substrate since poly-lysine is a nonspecific attachment factor for cells; however, certain cells can digest poly-lysine. In this case, poly-D-lysine should be used as the attachment factor so that the cells are not disrupted by excessive uptake of L-lysine.
Other uses for poly-lysine have been reported as well:
• Conjugation to methotrexate for increased drug transport.
• Microencapsulation of islets.
• Use in simple reproducible procedure for chromosomal preparations from a variety of tissues.
• Immobilation of living cells in biocompatible semipermeable microcapsules.
• In the preparation of polycationic beads.
• Conjugation to albumin and horseradish peroxidase to enhance cellular uptake.
Poly-D-lysine hydrobromide is a synthetic amino acid that enhances cell adhesion to solid substrates. It has additionally been shown to eliminate prion proteins from infected cells.
