Description: This MAb reacts specifically with heat shock protein HSP27 in human and monkey tissues and cell lines such as MCF-7. HSP27, also referred to as the Estrogen-Regulated 24K protein and HSP28, is one of several small heat shock proteins produced by all organisms studied. HSP27 synthesis is induced by elevated temperature, as well as by estrogen in hormone responsive cells. Interestingly, human HSP27 also shares greater than 50% homology with low molecular weight Drosophila HSPs and mammalian α-crystalline lens protein. Because of the estrogen responsive nature of HSP27, this protein has been studied extensively in human estrogen responsive tissues such as cervix, endometrium and breast tissue. Therefore HSP27 may be useful in classifying various hormone sensitive tumors.

Description: This gene encodes the homolog of the Drosophila melanogaster Nipped-B gene product and fungal Scc2-type sister chromatid cohesion proteins. The Drosophila protein facilitates enhancer-promoter communication of remote enhancers and plays a role in developmental regulation. It is also homologous to a family of chromosomal adherins with broad roles in sister chromatid cohesion, chromosome condensation, and DNA repair. The human protein has a bipartite nuclear targeting sequence and a putative HEAT repeat. Condensins, cohesins and other complexes with chromosome-related functions also contain HEAT repeats. Mutations in this gene result in Cornelia de Lange syndrome, a disorder characterised by dysmorphic facial features, growth delay, limb reduction defects, and mental retardation. Two transcript variants encoding different isoforms have been found for this gene.

Description: HEATR3 is a 680 amino acid protein existing as three alternatively spliced isoforms and containing two HEAT (Huntingtin, elongation factor 3 (EF3), protein phosphatase 2A (PP2A) and the yeast PI3-kinase Tor1) repeats. HEAT repeats form rod-like helical structures that are involved in intracellular transport. HEATR3 is encoded by a gene located on human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The GAN gene is located on chromosome 16 and, with mutation, may lead to giant axonal neuropathy, a nervous system disorder characterised by increasing malfunction with growth. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, a gastrointestinal inflammatory condition.

Description: Recognizes a 60 kDa protein, identified as the heat shock protein 60 (hsp60). Its epitope is localized between aa 383-447 of human hsp60. A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp), more appropriately called as stress response proteins (srp). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone LK1, unlike LK2, recognizes only the mammalian (not bacterial) hsp60 and is useful in distinguishing hsp60 from mammals and bacteria.

Description: PIRT is a 137 amino acid multi-pass membrane protein. Highly conserved among vertebrates, PIRT consists of two transmembrane domains and one putative C-terminal phosphoinositide-binding domain. Although PIRT is expressed in peripheral nervous system, with highest levels in dorsal root ganglion and trigeminal neurons, and lowest levels in sympathetic and enteric neurons, it is not expressed in spinal cord. PIRT is a required component of the VR1 complex, which positively regulates VR1, a sensor of both noxious heat and capsaicin. Correspondingly, PIRT knockout results in impaired responses to noxious heat and capsaicin exposure, while VR1 remains unaltered. The gene that encodes PIRT maps to human chromosome 17p13.1.
